How To Make Lyophilization Buffer

In order to freeze dry a cell culture of yeast, bacteria or fungus, it is important to use the right media. This recipe is for lyophilization buffer, for use in preserving your cell lines for long term storage by freeze drying.

Difficulty: Easy
Time Required: 1 hour

Here's How:

  1. Mix together in 1 L deizonized water: 5 g dextran (0.5%), 75 g sucrose (7.5%) and 10 g sodium glutamate (1%).

  2. Autoclave the 1 L mixture and allow it to cool before applying to culture plates during the lyophilization procedure.

What You Need:

  • One litre bottle or flask
  • Deionized water
  • Dextran
  • Sucrose
  • Sodium glutamate
  • Autoclave
